- Indian Innovation: Indian researchers have developed RK-251, an experimental “smart” cancer drug designed to become active mainly inside cancer cells.
- Developed by: Collaborative research led by scientists from Institute of Advanced Study in Science and Technology (IASST) and IIT-Guwahati.
- Target: It has shown promising activity against triple-negative breast cancer (TNBC) in preclinical studies.
How Does RK-251 Work?
- Prodrug: RK-251 is designed as a prodrug, meaning it becomes pharmacologically active after a specific trigger.
- Trigger — ROS: Cancer cells often have higher levels of Reactive Oxygen Species (ROS) than normal cells.
- Selective Activation: RK-251 responds to the high-ROS environment inside cancer cells.
- Release of NBDHEX: Once activated, it releases NBDHEX, an anticancer compound.
- Target: NBDHEX inhibits GSTP1, a protein associated with cancer-cell survival and resistance mechanisms.
- Objective: The approach aims to reduce damage to healthy cells compared with conventional chemotherapy.
Theranostic Feature
- Fluorescent Component: RK-251 incorporates a near-infrared fluorescent component.
- Dual Function: It combines therapy + diagnostic monitoring, a concept known as a theranostic approach.
- Activation Signal: Fluorescence can indicate activation of the drug in an ROS-rich environment.
Preclinical Findings
- Triple-Negative Breast Cancer: RK-251 showed strong activity against aggressive TNBC cells in laboratory studies.
- Healthy Cells: It showed substantially lower effects on healthy cells under the tested conditions.
- Zebrafish Model: Studies using zebrafish embryos (Danio rerio) found no obvious signs of acute toxicity and demonstrated the expected fluorescence response.
- Current Status: Preclinical stage — it has not yet been established as a human treatment and requires further safety and efficacy testing.
Exam Trap: RK-251 is not yet a proven replacement for chemotherapy. It is an experimental candidate undergoing preclinical evaluation.
